High sugar intake linked to low dopamine release in insulin resistant patients

June 10, 2013 ? Using positron emission tomography (PET) imaging of the brain, researchers have identified a sweet spot that operates in a disorderly way when simple sugars are introduced to people with insulin resistance, a precursor to type 2 diabetes. For those who have the metabolic syndrome, a sugar drink resulted in a lower-than-normal release of the chemical dopamine in a major pleasure center of the brain. This chemical response may be indicative of a deficient reward system, which could potentially be setting the stage for insulin resistance. This research could revolutionize the medical community's understanding of how food-reward signaling contributes to obesity, according to a study presented at the Society of Nuclear Medicine and Molecular Imaging's 2013 Annual Meeting.

"Insulin resistance is a significant contributor to obesity and diabetes," said Gene-Jack Wang, MD, lead author of the study and Professor of Radiology at Stony Brook University and researcher at the U.S. Department of Energy's Brookhaven National Laboratory in Upton, N.Y. "A better understanding of the cerebral mechanisms underlying abnormal eating behaviors with insulin resistance would help in the development of interventions to counteract the deterioration caused by overeating and subsequent obesity. We suggest that insulin resistance and its association with less dopamine release in a central brain reward region might promote overeating to compensate for this deficit."

An estimated one-third of Americans are obese, according to the U.S. Centers for Disease Control and Prevention. The American Diabetes Association estimates that about 26 million Americans are living with diabetes and another 79 million are thought to be prediabetic, including those with insulin resistance.

The tendency to overeat may be caused by a complex biochemical relationship, as evidenced by preliminary research with rodents. Dr. Wang's research marks the first clinical study of its kind with human subjects.

"Animal studies indicated that increased insulin resistance precedes the lack of control associated with pathological overeating," said Wang. "They also showed that sugar ingestion releases dopamine in brain regions associated with reward. However, the central mechanism that contributes to insulin resistance, pathological eating and weight gain is unknown."

He continued, "In this study we were able to confirm an abnormal dopamine response to glucose ingestion in the nucleus accumbens, where much of the brain's reward circuitry is located. This may be the link we have been looking for between insulin resistance and obesity. To test this, we gave a glucose drink to an insulin-sensitive control group and an insulin-resistant group of individuals and we compared the release of dopamine in the brain reward center using PET."

In this study, a total of 19 participants-including 11 healthy controls and eight insulin-resistant subjects-consumed a glucose drink and, on a separate day, an artificially sweetened drink containing sucralose. After each drink, PET imaging with C-11 raclopride-which binds to dopamine receptors-was performed. Researchers mapped lit-up areas of the brain and then gauged striatal dopamine receptor availability (which is inversely related to the amount of natural dopamine present in the brain). These results were matched with an evaluation in which patients were asked to document their eating behavior to assess any abnormal patterns in their day-to-day lives. Results showed agreement in receptor availability between insulin-resistant and healthy controls after ingestion of sucralose. However, after patients drank the sugary glucose, those who were insulin-resistant and had signs of disorderly eating were found to have remarkably lower natural dopamine release in response to glucose ingestion when compared with the insulin-sensitive control subjects.

"This study could help develop interventions, i.e., medication and lifestyle modification, for early-stage insulin-resistant subjects to counteract the deterioration that leads to obesity and/or diabetes," said Wang. "The findings set a path for future clinical studies using molecular imaging methods to assess the link of peripheral hormones with brain neurotransmitter systems and their association with eating behaviors."

Kanye West debuts new songs at Governors Ball

NEW YORK (AP) ? Kanye West closed the three-day Governors Ball on New York's Randall's Island with a set that featured his familiar hits as well as a batch of new, darker tracks from his upcoming album.

West kicked off his set Sunday night with the song "Black Skinhead," with flashing visuals in the background, including the words "not for sale." He later performed "New Slaves." Both tracks will appear on "Yeezus," his sixth album, which comes out June 18.

The crowd responded nicely to some of those songs, but it was lukewarm compared with West's performances of his well-known hits, including "Stronger," ''Good Life," ''Jesus Walks" and "All Falls Down."

Guns N' Roses headlined Governors Ball on Saturday. Kings of Leon was to headline Friday but pushed their performance to Saturday because of the bad weather.

(AP) ? Cicely Tyson, who returned to Broadway this season for the first time in 30 years, has taken home the Tony Award for best leading actress in a play.

In Horton Foote's "The Trip to Bountiful," Tyson plays a widow in 1953 whose only desire is to revisit her old home in Bountiful and recapture purpose she lost when she left for Houston.

Tyson, 88, beat out Laurie Metcalf, Amy Morton, Kristine Nielsen and Holland Taylor.

Tyson was nominated for an Oscar for her role in "Sounder" in 1972. Other film credits include "Fried Green Tomatoes," ''Bustin' Loose" and "Jefferson in Paris."

She won Emmys for "The Autobiography of Miss Jane Pittman" and "Oldest Living Confederate Widow Tells All." She was also nominated for her role in the miniseries "Roots."

Carbon nanotubes for molecular magnetic resonances

June 9, 2013 ? More resistant than steel, carbon nanotubes are one of the strongest and hardest materials known. Their impressive electrical and thermal properties make them an extremely versatile material. Hollow on the inside and only one-atom thick, they lend themselves to a large variety of potential uses, from tennis rackets and bulletproof vests, to electronic components and energy storage devices. New research shows that they may also hold the potential for revolutionizing medical research with magnetic resonance imaging of individual molecules.

Scientists from ICFO- Institute of Photonic Science, in collaboration with researchers from the Catalan Institute of Nanotechnology (ICN2) and the University of Michigan, have been able to measure weak forces with sensitivity 50 times higher than what has been achieved to date. This significant improvement represents a turning point in measuring very weak forces and opens the door for magnetic resonance imaging at the molecular scale. Dr. Adrian Bachtold, who began this research at the Catalan Institute of Nanotechnology before transferring his research group to ICFO, explains in an article published in Nature Nanotechnology that they were able to prepare the carbon nanotubes to act as probes that vibrate with an intensity proportional to an electrostatic force. With the use of ultra-low-noise electronics, the group led by Bachtold was able to measure the amplitude of the vibration of these nanotubes and thus surmise the intensity of the electrostatic force.

"Carbon nanotubes are similar to guitar strings which vibrate in response to the force applied. However, in the case of our experiment, the forces that cause the vibration are extremely small, similar to the gravitational force created between two people 4500 km apart," explains Bachtold. In the last ten years scientists have made only modest improvements in the sensitivity of the measurement of very weak forces. This new discovery marks a before and after and points to carbon nanotubes playing an important role in future technologies for MRIs of individual molecules.

Conventional magnetic resonance imaging registers the spin of atomic nuclei throughout our bodies which have been previously excited by an external electromagnetic field. Based on the global response of all atoms, it is possible to monitor and diagnose the evolution of certain diseases. However, this conventional diagnostic technique has a resolution of a few millimeters. Smaller objects have an insufficient total number of atoms to allow for the observation of the response signals.

"The results presented are very promising for measuring the force created by each individual atom and consequently its spin. In the future this technique could revolutionize medical imaging" concludes Bachtold.

(AP) ? Cher has a date with NBC's "The Voice."

NBC said Monday the pop star will perform live on the talent contest's June 18 season finale.

She will sing "Woman's World," the first single off her upcoming album of the same name, the network said.

Cher, who turned 67 in May, joins other pop artists who have appeared on NBC's hit series and other TV contests to promote new records. They include "Voice" guest stars Rod Stewart and Lady Antebellum.

Cher's "Woman's World" album, her first in 12 years, is due out later this year.

The Grammy, Oscar and Emmy-winner has scored a No. 1 single on the Billboard pop charts in each of the last six decades, including her 1998 single "Believe."

Brother-in-law of Chinese Nobel winner jailed for 11 years

HUAIROU, China (Reuters) - A Chinese court on Sunday sentenced the brother-in-law of jailed Nobel Peace Prize laureate Liu Xiaobo to 11 years in prison on charges of fraud in a case that rights activists have called another example of official retribution on the Liu family.

Supporters of Liu Hui say his case was trumped up, aimed at thwarting the increasing attention by the rights community on the plight of Liu Xia, who has remained under effective house arrest since her husband Liu Xiaobo won the Nobel Prize in 2010.

The court in Huairou, a one-hour drive northeast of Beijing, convicted Liu Hui, a manager in a real estate company in the southern city of Shenzhen, on charges of defrauding a man called Zhang Bing of 3 million yuan ($490,000) with another colleague, lawyer Mo Shaoping told reporters.

"As Liu Hui's defense attorney, I definitely do not approve of this verdict, because we see this fundamentally as a civil issue, and it fundamentally does not constitute criminal fraud. Also, there is not sufficient evidence," Mo said.

Liu Hui has maintained his innocence, according to his lawyers.

In a rare statement to media, a weeping Liu Xia told reporters from the front passenger seat of her car as she drove away from the courthouse that she was extremely angry with the verdict and vowed to launch an appeal.

"I absolutely cannot accept this. This is simply persecution," she said. "This is completely an illegal verdict."

Liu Xia said she had "completely lost hope" in the government.

"I can't even leave my house."

After about two minutes, security forced journalists away from the car, which moved off.

Liu Hui was out on bail last September, but then arrested again in January, after several rights activists and foreign reporters forced their way past security guards late last year to visit Liu Xia, one of his other lawyers, Shang Baojun, told Reuters before the verdict.

Raphael Droszewski, a first secretary at the European Union Delegation to China, told reporters outside the courthouse that the EU was deeply concerned by Liu Hui's sentence. He called for Liu Xiaobo's release and the ending of restrictions on Liu Xia.

"As it is clear Liu Xia is under house arrest, the EU urges the Chinese authorities to end all forms of extra legal restriction on her," he said.

SETBACK FOR REFORM

The ruling is seen as a setback for hopes for political reform from new Chinese President Xi Jinping, whose government has detained anti-corruption activists and tightened free expression on the Internet following his appointment in March.

"Everything related to the Liu Xiaobo case previously could have been seen as the legacy of the previous leadership," said Nicholas Bequelin, a researcher at Human Rights Watch, a New York-based advocacy group.

"But with this sentence the new leadership buys into this suppression and persecution of the family. It deems the prospect unlikely that the government is amenable to the release of Liu Xiaobo, or will make any concessions on the case."

The case will also renew international criticism of the plight of Liu Xiaobo's family.

The verdict was handed down within hours of Xi and U.S. President Barack Obama completing an informal summit in California. U.S. and European diplomats were present outside the courthouse in Huairou, a one-hour drive northeast of Beijing.

Speaking in California after the summit, China's top diplomat, State Councilor Yang Jiechi defended the country's achievements in human rights as "there for all to see".

"With China's economic and social development, human rights causes in China will enjoy even greater progress in the future," Yang told reporters, though he did not refer directly to the Liu Hui trial.

Liu Xiaobo, a veteran dissident involved in the 1989 Tiananmen Square pro-democracy protests crushed by the Chinese army, was jailed for 11 years in 2009 on subversion charges for organizing a petition urging the overthrow of one-party rule.

His wife Liu Xia is rarely allowed out and is almost never allowed to receive visitors. She has not been convicted of any crime.
